Endotoxin, Sterility and the Limits of “Research Grade”
“Research grade” is a commercial description, not a defined standard. Knowing what it does not include is more useful than knowing what it does.
What a standard certificate covers
A typical research peptide certificate reports identity and chromatographic purity, and sometimes net peptide content or water. Purity in this context means peptide-related purity: the proportion of peptide material that is the target sequence. It says nothing about anything that is not a peptide.
What it usually does not cover
Three categories fall outside a standard purity analysis, and each matters in different work.
Endotoxin
Bacterial lipopolysaccharide is not detected by RP-HPLC and does not appear in a purity figure. It is biologically active at very low concentrations and is a well-documented confounder in cell culture, particularly in immune and inflammatory readouts, where it can produce responses mistaken for a compound effect. Sterility
Lyophilised peptides are not sterile products unless specifically produced and tested as such. A vial can be low in endotoxin and still carry viable organisms, and the two are separate tests answering separate questions.
Residual solvents and reagents
Synthesis and purification leave traces — scavengers, coupling reagents, organic solvents. These are generally present at low levels and generally irrelevant, but they are not zero and they are not covered by a peptide purity figure.
When to ask for more
For biochemical and analytical work, standard identity and purity data is usually sufficient. For cell culture — especially immune, inflammatory or cytokine readouts — endotoxin data is worth requesting, and a vehicle control that has been through the same handling is worth running regardless.
If a supplier cannot say whether a batch was tested for endotoxin, the answer is that it was not. That is not necessarily disqualifying; it is information you need in order to design controls. Related: choosing a research peptide supplier and why certificates disagree.
