Weighing Lyophilised Peptides: Hygroscopicity and Static

Weighing a lyophilised peptide accurately is harder than it looks, and the errors are systematic rather than random — they push in one direction and quietly bias every concentration downstream.

Water uptake

Lyophilised peptides are hygroscopic. A vial opened at ambient humidity begins absorbing atmospheric water immediately, and a light, fluffy cake presents a large surface area to do it with. Mass measured after several minutes on an open balance can differ appreciably from mass at the moment of opening.

The standard mitigation is to allow the vial to equilibrate to room temperature before opening — opening a cold vial condenses moisture directly onto the contents — and to work quickly, ideally in a low-humidity environment or a glove box for demanding applications.

Static charge

Dry peptide powder holds static charge readily. On a microbalance this produces drift, and physically it causes powder to jump to the spatula, the weighing boat or the vial wall. For milligram quantities the loss can be a meaningful percentage. An antistatic device near the balance, or metal rather than plastic weighing tools, addresses most of it.

The alternative: weigh nothing

The most robust approach for many workflows is to avoid weighing altogether. Reconstitute the entire vial with a known solvent volume, then work from that stock by dilution. This removes both the hygroscopicity and static problems in one step, and it makes the vial’s stated content the only quantity you need to trust.

Residual solvent and water in the certificate

Some certificates report water content, commonly by Karl Fischer titration, and residual solvent separately. Where those figures are available they let you correct a gross weight toward actual peptide mass. Where they are not, net peptide content by amino acid analysis or quantitative nitrogen determination is the more direct figure to look for.

Practical checklist

Equilibrate the vial to room temperature before opening. Keep exposure to open air short. Use antistatic precautions. Prefer whole-vial reconstitution over partial weighing where the experiment allows. Record ambient humidity if you are troubleshooting inconsistent results between sessions — it is a more common culprit than it is given credit for.
